ABSTRACT

Objective To investigate the polymorphisms of the 19 Y-STR loci in the Han population in Northeast China, and analyze the genetic relationships of 15 ethnic groups in northeast Asia region, and to evaluate their forensic value and population genetic value. Methods The 19 Y-STR unrelated Han males in 200 unrelated Han males in Northeast China were amplified with the Y-filer system, and the PCR products were analyzed by the 310 Genetic Analyzer. The AMOVA analysis, cluster analysis and MDS analysis were calculated by Arelquin3.11,Mega4.1 and SPSS17.0.Results The polymorphisms of 19 Y-STR loci in the Han population in Northeast China had generally higher gene diversity value which were ranged from 0.350 9 (DYS391) to 0.971 1 (DYS385a/b),and totally 200 haplotypes were observed. The 19 Y-STR loci displayed high genetic polymorphisms in the Han population in Northeast China, indicating that these 19 loci were useful genetic markers for forensic personal identification and paternity testing. There were distinctions among 15 ethnic groups. The genetic distance between 15 ethnic groups were ranged from 0.000 9 to 0.643 2,and the conclusion of cluster analysis and MDS analysis were similar to the ethnogeny research and ethnic migration history. Conclusion The 19 Y-STR loci displayed high genetic polymorphisms in the Han population in Northeast China, and these 19 loci were useful genetic markers for forensic personal identification and paternity testing.

ABSTRACT

AIM:To investigate the effects and mechanisms of sphingosine-1-phosphate receptor-2 (S1P2R) on lipopolysaccharide (LPS)-induced acute lung injury (ALI).METHODS:ALI model was induced by intratracheal ad-ministration of LPS in both wild-type mice and S1P2R-deficient mice. The pathological changes in the lung tissues were ob-served, and the protein concentration , total cell number, neutrophil ratio, TNF-αlevel and IL-6 level were determined in the bronchoalveolar lavage fluid (BALF) 24 h after LPS injection. In order to investigate the mechanisms of S1P2R in LPS-induced ALI, 10 min before LPS injection, both wild-type mice and S1P2R-deficient mice were injected with nitric oxide synthase inhibitor by tail vein injection , the pathological changes of the lung tissues were observed , and the protein concen-tration and total cell number in BALF were determined 12 h after LPS injection .RESULTS: Compared with wild-type mice, S1P2R-deficient mice showed more severe LPS-induced ALI, and the protein concentration , neutrophils and inflam-matory cytokines in BALF were significantly increased in S1P2R-deficient mice.Administration of nitric oxide synthase in-hibitor Nω-L-nitro-arginine methyl ester protected S1P2R-deficient mice from aggravation of ALI .CONCLUSION:S1P2R mediates the protection from LPS-induced ALI possibly through inhibiting nitric oxide synthase .

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the mitochondrial DNA (mtDNA) five coding region sequence polymorphisms encompassing positions nt3954-4506, nt5218-5974, nt7942-8711, nt10296-10653, and nt14496-14867 in Chinese Han population of Yanbian area, Jilin province.</p><p><b>METHODS</b>Polymerase chain reaction (PCR) and direct sequencing method were used to detect the haplotype distribution of mtDNA coding region in 200 unrelated Chinese Han individuals.</p><p><b>RESULTS</b>One hundred and ten haplotypes were observed in the 200 individuals. The gene diversity was 0.9879 and the random match probability was 0.0171. Compared with the Anderson's sequence,81 nucleotide variants were obtained,of which 66 were previously registered in MITOMAP,and 15 were novel.</p><p><b>CONCLUSION</b>The obtained data suggest that these sequence polymorphisms are valuable genetic markers for personal identification when added to mtDAN control region investigation, and thus could be used as basic data for the forensic application in Chinese Han population.</p>

ABSTRACT

Objective To find out the differences on the knowledge, attitude and behavior related to Hepatitis B transmission between Hepatitis B patients and control group( healthy people) and to analyzing the risk factors of Hepatitis B transmission. Method Adopted case-control study, 50 people who were Hepatitis B patients and 100 people as control group were selected to conduct retrospective investigation. Results Skin wound, disqualified medical treatment, unhealthy habits and manners are high risk factors to Hepatitis B transmission. The knowledge, attitude and behavior of Hepatitis B are very important to prevent Hepatitis B transmission. Conclusion There was a misunderstanding on the knowledge about Hepatitis B transmission among Hepatitis B patients. They thought daily contact was the key way of HBV transmission. They underestimated the blood transmission and sexual transmission. We need to conduct more propaganda on the knowledge of Hepatitis B prevention to change peoples' unhealthy habits and behavior.
